What the human evidence actually shows

Here is where honesty matters most. The direct human research on PQQ is limited, and the studies that exist are small and short.

One frequently cited trial gave adults 20 mg of PQQ per day for eight weeks and reported improvements across measures of fatigue, tension, mood, and several aspects of sleep, including how easily people fell asleep and how rested they felt on waking (Nakano et al., 2012). Those results sound encouraging, but the study included only around seventeen participants, ran for a matter of weeks, and relied heavily on subjective questionnaires, which are prone to placebo effects in a small group.

On the mechanistic side, a human crossover study found that supplementing PQQ shifted certain blood markers of inflammation and produced changes in urinary metabolites linked to mitochondrial activity (Harris et al., 2013). This is useful because it shows PQQ does something measurable in the human body, but changes in biomarkers are not the same as feeling more energetic or performing better, and the study was again very small, with roughly ten subjects.

For the brain, the most-cited work is a randomized, placebo-controlled trial in older adults that reported improvements on some cognitive test scores with a branded PQQ disodium salt (Itoh et al., 2016). It is worth knowing that research on branded ingredients is often connected to the companies that sell them, which does not make it worthless but does warrant extra caution. More recent small trials in people with mild cognitive decline have reported changes in brain oxygenation and certain metabolic markers, but these too are early and modest.

The pattern across all of it is consistent: mechanistically interesting, human effects small, studies short and often industry-linked. PQQ is best described as promising and under-proven, not established.

Doses, safety, and the CoQ10 question

Most commercial PQQ products and most human studies land in the range of roughly 10 to 20 mg per day, usually taken once with food. There is no evidence that higher doses do more, and the safety data get thinner as the dose climbs.

On safety, the reassuring part is that in the short-term clinical studies conducted so far, PQQ at these doses has generally been well tolerated, and a European food-safety review concluded that PQQ disodium salt did not raise safety concerns at the intended intake levels (EFSA, 2017). The caveat is that “generally well tolerated in short studies” is not the same as proven safe for years of daily use, and the long-term data simply do not exist yet.
